What is TLC?

Thin Layer Chromatography (TLC) is a simple and powerful technique used to separate the components of a mixture. Like other types of chromatography, it works by exploiting the different affinities substances have for two phases:

  • Stationary phase – a thin layer of silica gel, alumina, or cellulose coated on a glass, metal, or plastic plate.

  • Mobile phase – a solvent that moves up the plate by capillary action.

As the solvent rises, it carries the mixture with it. Each component interacts differently with the stationary and mobile phases, causing them to separate.

How Separation Works

Polarity plays a major role in TLC. More polar compounds stick more tightly to the stationary phase and move slowly. Less polar compounds travel farther because they prefer the mobile phase. This difference in movement creates distinct spots on the plate, which can then be visualized and analyzed.

Why Use TLC?

TLC is widely used because it is quick, easy, and inexpensive. It works with organic compounds, inorganic compounds, and even biological mixtures. Chemists often use it to check reaction progress, identify compounds, or test purity.

Limitations of TLC

  • Quantifying results can be challenging

  • Very similar compounds may not separate well
